is it possible to use "gain" in this way.. first i put the gain value "100".. and i want it to change to "200" in 5 second.. ???any help please...

Do you plan to use the Clock block to determine when t>5secs? It is typically not possible to change a block parameter based on the output of another block. See http://blogs.mathworks.com/seth/2011/03/08/how-do-i-change-a-block-parameter-based-on-the-output-of-another-block/ for further explanation.

However, in your case, you can simply use a Product block instead of a Gain block. Use your input signal as one of the inputs, and for the other input connect a Switch block which takes "100" and "200" as its inputs - you can use a Clock block with a comparison operator to determine when to switch between the "100" and "200".
