Detecting shapes-lsm format?

I want to detect the number of blue coloured spots (nuclei to be precise) in an image of lsm format using matlab, however writing the code below is not detecting the shapes at all, instead as i do imshow a white page opens up.(Indicating that the entire picture has been converted into white) code: i=imread('01.lsm'); imshow(i) BW = im2bw(i, 0.9); BW = ~BW; imshow(BW)
Is it a problem with the code or with the format? Thanx in advance.

try looking at hist(i,100) and picking out a threshold manually. somewhere to the right of the big background spike on the left.

oh and imclearborder() bw=bwareaopen() and imfill(bw,'holes') could be pretty useful for you too
