.jpgのカラー画像をグレースケール化して、バイナリーイメージに変換後、regionpropsやimbinarizeで使用する方法はありますか？？

とりあえず、rgb2grayを試されてはいかがでしょうか。
imreadを使えばjpg画像も読めますので、
イメージのセグメンテーションと解析
等の例が参考になるかと思います。
