I need a help to find intial contour and final contour.

I have level set functions code in that there are four input 1.Input-input image(segmented image) 2.Iteration-number of iterations 3.initial contour and final contour. I have attached Brain MRI image,segmented image and level set function code.so i need a code to find initial contour and final contour for that input image and function call for level set functions .Can anyone help me..
