Regarding the Total Focusing Method-image reconstruction

I need to create an image based on the intensity values obtained from each pixel. First I need to create a grid so that it will represents the coordinate of pixel and grid should be based on the cross sectional area of specimen(60mmx25mm). What way I can create a grid so that the intensity values obtained from Matlab programming can can on those pixel to represent the image?
