int16 and fixed point transformation

Hi All! I am trying to build a process wich take place in a DSP . I want to evaluate with matlab.
I get uint16 data from the ADC. this data schould be filtered. the Filter works with fixed point (fract16).
should a transformation take place? How to do it?
